Description

The Calcium Kit II - Fluo 4 is a screening kit containing Fluo 4-AM, a reagent for the measurement of intracellular Ca2+, as well as buffers and other components necessary for the measurement.
Depending on the cell type and drugs to be added, the concentrations of Pluronic F-127 or Cremophor EL (solubilizing agents for Fluo 4-AM) and Probenecid (an inhibitor of anion transporters) can be adjusted as required.The kit includes a reagent that eliminates the need for conventional cell washing procedures by quenching the fluorescence of Fluo 4 present in the extracellular space. This allows for the rapid processing of a large number of samples, making the kit suitable for high-throughput screening (HTS).
However, depending on the cell type and added drugs, variations such as "high or low response" or "interactions with ligands" may occur. For measurements requiring minimal impact on the measurement system, we recommend using the sister product, "Calcium Kit - Fluo 4" (Wash Type).
The prepared Loading Buffer allows for the measurement of ten plates, each with 96 wells. For measurement, use a clear-bottom plate and a plate reader capable of downward excitation and fluorescence measurement.

Are there any differences in the measurement results between Wash and Non-Wash?

A

There is almost no difference.
However, since the non-wash type uses a quenching buffer, the rate of change in fluorescence intensity or response may differ depending on the cells or stimulants used.
